You're talking about the 100,000 BC humans, like Fortencho, correct? If so, then yes. The forerunners had knowledge of what they were doing though, as they were in the middle of war with the humans.

I think it was before the Human-Forerunner War. Forthencho destroyed Forerunner planets that were beginning to show signs of infection, which initiated the War.

From what I'd read, when Humans tested the Powder, they found nothing that suggested it would have any negative consequences long term.

Yep, as it was meant to be revenge against the forerunners, I doubt they knew of the source, either. Of course, LoA could probably blow everything I'm saying out of the water right now.
