Is having a fire team mandatory?
I am more of a solo gamer/lone wolf. So will the Destiny experience be hindered because I want to roll solo instead of with other players.

The mandatory ingredient for playing Destiny is the online connection. I generally like to lone wolf it too, save for certain situations. It'll be doable.
